imho there are 2 combos that truely shine for PvE
1. lock / lock and 2.mage / priest
2. there are many ways to play the lock / lock combo eg.
2 voidwalkers 10 mobs and 6 dots per mob.
1 voidwalker tanks maybe 3 - 5 mobs while the other lock uses an imp to improve DPS.
both locks have the ability to heal themselves and restore mana to a degree. if it all goes wrong you both have an instant self rez and nearly no down time. me and my girlfriend are currently running this set up to level our horde locks after switching from alliance where we both have high level chars. we have had no trouble with mobs 3-5 levels higher (altho considerably slower) and fly through all quests and grind mobs a couple of levels below us in almost laughable amounts of time.
2. the mage and priest combo imho is probably the quickest combo you can possibly use to grind from 1 - 60. this is doable as soon as the mage gains his AoE spells.
the mage simply rounds up say 10 mobs freezes them in place then spams AoE instants while the priest flash heals him. this method is SUPER quick and altho there is some down time involved it is considerably cheaper than other combos as the mage can make water and food.
another added bonus is the buffs seeing as priest can stam buff and mage can int buff.
as for PvP there are a hole number of combos that work well. generally speaking the standard approach is healer / DPS.
one word about healers however is paladin is NOT the best healer for 2's and 3's the simple reason being is both there healing spells are timed casts which means that they use a cast bar. any good team with a dispeller will be able to dispell or interupt any of the paladins heals as soon as they see the cast bar, this significantly reduces a paladins ability to main heal in the smaller PvP brackets. and there only instant heal (only available if the paladin is specced in the holy tree) is laughable at best considering the way it drains mana.
prefered flavor of the month healer would have to be druid for HoTs.
as for DPS arms warrior is a solid choice but susceptible to CC. paladin since patch 2.4 has good burst potential but if you go OOM your dead. rogue is a solid choice with good CC but lower survivailty. ench shaman has good buffs. mage has good CC. warlock does insane damage and has the ability to self heal etc but needs to stay alive long enough for the DOTs to be effective. shadow priest has good DPS, the ability to heal to a degree, wont go OOM and can dispel etc etc.
other combos rely on bursting down the opposing team as soon as possible. teams like lock / lock. lock / rogue. lock / shadow priest are all viable but against heavily armored classes or classes with good survivabilty you have your work cut out for you.
my recomendations for 2's in arena would be
lock / priest
lock / lock
druid / warrior and
druid / rogue

This is the very best duo imo for pve, have used all duo combos with different specs and nothing comes close to this one. This is a hybrid combo that works for everything.
Paladin-Shockadin spec (holy/Ret)
Skills- Mining/Blacksmithing: Very High Income
Mount-Free
Abilities-DPS, Heal, Aggro,party buffs,reserection,tank
Mage-Eternal Flame spec (Arcane/Fire)
Skills-Herbalisim/Alchamy
Mount-paid for by paladin's high income
Abilities-Very high DPS, never ending mana, AoE, C.C. intelect buff for paladin, group teleport.
With this combo, my wife and I have no down time, neither of us ever run out of mana, her income can buy anything we need out of the A.H., the paladin can greatly inhance the mages' abilities with the buff's, and with the aggro redirect; the mage never gets hit. This type duo covers everything because we picked characters, specs, and skills that would either compliment the other's, or fill in the holes the other one lacked.
No competition for any gear due to niether of us use even similar gear. You can run most instances with just about one of anyother class player, or one of the same type too. The DPS is off the charts because of the ret/fire aspect and the holy/alchamy/arcane keeps mana and health at 100% at all times. In the rare event that the mage gets into trouble, ice block and paladin pulls the mob with aggro. Pally pulls the mob a good distance off, then mage melts face.....end of story. Works in crowds too. Same idea, but use ice shards and blizzard, ice block.....pally pulls again....mage uses arcane shield and repeat.
Done, everyone is dead but mage and pally. Eat, drink, make potions and go get some more. With a real duo....not just a catch ya when your on kinda thing, you have to build a team that works where one can do what the other cant. Now on the flip side, the mage can survive as long as you dont pull more than one mob. The potion cooldown will eventualy get you killed. The paladin can hold their own till the mana runs out....then it's game over. But with all that said, this is the very best tag-team in the game.

For grinding there are two amazing combos.
Discipline Priest+Mage
(Reflective Shield+Power Infusion+Aoe=hax)
Or Warlock+Felguard

Best duo for PvP IMO: Shadowpriest or 'lock for DPS & Shadow/Holy priest for healing. GG
I don't have much experience in PVE so your guess is as good as mine =P

Yeah, you're thinking "What?! Those aren't classes!" That's right, it's my buddy Dan, out in Seattle, and me, up here in Saskatoon. We've each got a stable of 60's, and duo together with any 2 of them. We've gotten to the point where we understand each other's gameplay so well, that we predict each other's moves and act accordingly.
So really, I think if you can find someone you play well with and get along with, you'll grind really well.
We've tried warlock/pally, warrior/priest, hunter/shaman, rogue/druid, and pretty much every other combination with those classes.. We just don't slow down for anything.

best duo for pvp?
Warlock + Warlock. Affliction specced = uberGG.
Heck, I soloed in the arena vs a pally/rogue, and I still managed to win. did 16k damage too =) (pally shielded and healed himself and rogue to full.) (as a Aff lock)

For the PvP Arena 2v2 system, I can guarantee you that a majority of the duos on the top of the ladder will contain 1 of the following 2 classes. Paladin or a Warlock. The two together would make a very strong combination. In fact, a Paladin with any other class (preferably high DPS) is going to be very hard to beat.
My personal favorite for 2v2 Arena...Druid/Warlock.

For PVE leveling, Druid and Rogue. Very versitile.
Tank/DPS
Healer/DPS
DPS/DPS
and can stealth past mobs to speed up certain quests/quest lines.
Me and my friend's mains (starting way back when) were/are a rouge and druid and even though it was the first time either of us had played an MMO, we burned through to 60.

At the moment, two Warlocks. Seriously. One need only see the damage they do in 2v2 Arena right now to swear off entering it entirely.
In BC, probably a well-geared hybrid and DPS. The hybrid could be used to heal the DPS, and if things get hairy can switch into DPS themselves. Plus, Paladins and Druids right now are doing some scary damage.

A friend and myself run a Shadow Priest/Aff Lock combination. We are both 49, so its not the same as 60. But we can hold our own against anybody in the 2v2 arena and BGs.

Currently My buddy and I have lvld up since lvl 10 together. Weve solod every instance so far and do pretty much all quests just the 2 of us.
I'm a lvl 42 druid now. He is a lvl 43 warrior.
We just finished soloing all of SM at 41 druid and 42 warrior.
I heal while he kills. I also help kill and step out to heal when needed.
PvP works about the same. I can hold off horde long enough to allow him to deal dmg and I can heal him and myself.
Once we hit 70 and start going into arenas I cant see us being beat.

I find when I group up with my friend, we slaughter through things amazingfly fast
Fury spec'd Warrior
Marksman spec'd Hunter
He holds the aggro while my pet and my fury of arrows cuts them to pieces

I'd say Pally or Druid with a Rogue. The utility of either, plus a decent stunlock, can keep most things still long enough to die... and if the Rogue is taken out, a combat rez goes a long way.

My girlfriend and I have had an absolute blast playing our Druid Duo. I tank/dps (feral spec) while she heals/dps (resto spec). Im a skinner/leatherworker, she's a tailor/enchanter. We stealth pasts mobs we dont want to fight (or to go back to get a quest she missed in a dungeon). I can tank or DPS PUGs (feral gear is pretty versatile for bear/kitty forms when you get the right gear), while she heals very well. We're mid 50's now and haven't PvPd yet, but we were looking to start doing that soon (I think its going to work well).
I like it better than the druid/rogue because bear form allows us to do pretty much any content in the game. I think we're going to try a paladin/priest combo when the expansion comes out.

Warlock/Priest is the easiest time my wife and I have yet had in the game. We've duo'd hunter/druid, paly/priest, rogue/priest, druid/druid...none of them can stack up to the ease of the entire game 1-60 that a lock (preferably affliction) and a priest (pref shadow :P), can handle. The entire game has seriously been an absolute breeze with that combo.

Me and my boyfriend are a rogue/druid team as well.
Being able to stealth together past mobs is awesome. Our dps is great together, nothing lives long. When there are alot of mobs, he just goes bear. He barely (lol bearly) has to even heal us with his imp feral skill. We can't wait to try out our dynamic in BC.
And let me tell you, grouping with a druid is always a good idea when it comes to doing dungeons. The fact they can become a healer/tank/dps based on what's needed helps you get into groups much easier.

Lock and druid. The slow sustained healing makes lock mana last "5 eva". The dps is nice and there is a great versatility in tanking/DPS roles. Probably the most enjoyable experience i've had was duo-ing with a moonkin.

Any class that can hold aggro (tank) + a healing class (healer)
Tanks: Warrior, Paladin, Feral Druid, Shadow Priest, Shaman, Warlock, Hunter
Healer: Paladin, Druid, Holy Priest, Shaman
PVP

DPS class that is good versus any other class + another of the same type. Rogue and Mage is probably the best combo since these two are the highest damage dealers.
