While the glyph removes the CD on misdirect, since the skill's duration is 4 seconds, I'm willing to bet that there's an internal CD effect that won't let you re-cast misdirect while the first one is still active. You shouldn't need to cast 2 misdirects within 4 seconds of each other, because during those 4 seconds all your threat is getting misdirected anyway.
