When I used to play Dark Age of Camelot I was all over it, I was an officer in a big guild for a while and later the GM of another. The key was simply to have regular time to play and a willingness to do some people-herding when it becomes necessary.

Given that I primarily came to WoW because it's much more rewarding to play casually, I can't say I can see myself taking on responsibilities of running a significant guild again.
